To Throw Punches: Go to Options, Controllers and change the desired combination of buttons to use in the game.

How To Set Rumble Attack: Options, Controller, Set buttons to High Block, Low Block. Push together to activate the Rumble Attack and to regain energy while the opposition is down.

Activate Rumble Attack: (Q) + (E) 

Use Rumble Attack: (Z) + (X) Hold together and use the arrow keys for different combos or Toggle Left, Right quickly. Up Down arrows can be used for different combos.
